WGA signal and membrane outlines

WGA labels accessible glycans. Check outline continuity for segmentation; intensity is not a direct measure of membrane amount.

Dashed line: noncovalent target recognition. Shapes are schematic. AF marker: generic dye symbol.

Understand the staining signal

WGA recognizes specific glycans, so signal depends on glycosylation and accessibility. Brightness differences between cells do not directly measure membrane amount. For outline detection, first check continuity and separation of adjacent cells.

Check segmentation bias

Review bright, dim and differently shaped cells for missed outlines, merged cells and intracellular signal assigned to boundaries. Use consistent analysis rules across groups and record exclusions and their reasons.
