Formulas
Cell conc. = (cells counted / squares) × dilution factor × 10⁴
10⁴ = conversion factor from 0.1 mm³ to 1 mL
Viability (%) = live cells / total cells × 100
Operating tips
- Mix cell suspension 1:1 with trypan blue (2× dilution) and count live/dead cells.
- Count all 4 large squares and average to reduce error.
- Each large square has a volume of 0.1 mm³ (10⁻⁴ mL).
- Optimal density: 20–50 cells per square; dilute further if more crowded.
- Complete counting within 3–5 min after staining to avoid dye toxicity.
