About common buffers
PBS (Phosphate-buffered saline)
The most widely used isotonic buffer for cell washing, dilutions, and immunological assays. pH 7.4 is isotonic with physiological conditions.
TBS (Tris-buffered saline)
Tris-based buffer system commonly used for membrane washing and antibody dilution in Western blot. Adding 0.1% Tween-20 gives TBST.
Tris-HCl
Versatile biological buffer, pH range 7.0–9.0. 1 M stock is standard; note temperature effect on pKa (–0.03 pH/°C).
EDTA
Metal ion chelator that inhibits metalloprotease activity. 0.5 M pH 8.0 stock is commonly used in TE buffer preparation.
